As the pregnancy progresses, increase in the oestrogen concentration promotes uterine contractions. These uterine contractions facilitate moulding of the foetus and downward movement of the foetus. The descent of the foetus causes dilation of cervix of the uterus and vaginal canal resulting in a neurohumoral reflex called Foetal ejection reflex or Ferguson reflex. This initiates the secretion of oxytocin from the neurohypophysis which in turn brings about the powerful contraction of the uterine muscles and leads to the expulsion of the baby through the birth canal.

(ii) The immune response of the body is stimulated by a DNA molecule.
(iii) A DNA vaccine consists of a gene encoding an antigen protein, inserted on to a plasmid, and then incorporated into the cells in a target animal.
(iv) DNA instructs the cells to make antigenic molecules which are displayed on its surfaces. This would evoke an antibody response to the free floating antigen secreted by the cells.
(v) The DNA vaccine cannot cause the disease as it contains only copies of a few of its genes.
(vi) DNA vaccines are relatively easy and inexpensive to design and produce.
(vii) Vaccines produced by these new techniques have definite advantages like producing target proteins, long lasting immunity and trigger immune response only against specific pathogens with less toxic effects.

The primary endosperm nucleus divides immediately after fertilization. It develops into an endosperm which is a nutritive tissue and a regulatory structure. The zygote divides only after that to form an embryo so that it can get nourishment from the endosperm that has formed earlier.

According to the theory of chemical evolution, primitive organisms in the primordial environment of the Earth evolved spontaneously from inorganic substances and physical forces such as lightning, UV radiations, volcanic activities, etc. Oparin (1924) suggested that the organic compounds could have undergone a series of reactions leading to more complex molecules. He proposed that the molecules formed colloidal aggregates or 'coacervates' in an aqueous environment. The coacervates were able to absorb and assimilate organic compounds from the environment. Haldane (1929) proposed that the primordial sea served as a vast chemical laboratory powered by solar energy. The atmosphere was oxygen free and the combination of CO2, NH3 and UV radiations gave rise to organic compounds. The sea became a 'hot' dilute soup containing large populations of organic monomers and polymers. They envisaged that groups of monomers and polymers acquired lipid membranes and further developed into the first living cell. Haldane coined the term prebiotic soup and this became the powerful symbol of the Oparin-Haldane view on the origin of life (1924-1929). Oparin and Haldane independently suggested that if the primitive atmosphere was reducing and if there was appropriate supply of energy such as lightning or UV light then a wide range of organic compounds can be synthesized.

PCR In Clinical Diagnosis: The specificity and sensitivity of PCR is useful for the diagnosis of inherited disorders (genetic diseases), viral diseases, bacterial diseases, etc., The diagnosis and treatment of a particular disease often requires identifying a particular pathogen. Traditional methods of identification involve culturing these organisms from clinical specimens and performing metabolic and other tests to identify them.
The concept behind PCR based diagnosis of infectious diseases is simple - if the pathogen is present in a clinical specimen its DNA will be present. Its DNA has unique sequences that can be detected by PCR, often using the clinical specimen (for example, blood, stool, spinal fluid, or sputum) in the PCR mixture. PCR is also employed in the prenatal diagnosis of inherited diseases by using chorionic villi samples or cells from amniocentesis. Diseases like sickle cell anemia, p-thalassemia and phenylketonuria can be detected by PCR in these samples. cDNA from PCR is a valuable tool for diagnosis and monitoring retroviral infections - egoTuberculosis by Mycobacterium tuberculosis.
Several virally induced cancers, like cervical cancer caused by Papilloma virus can be detected by peR. Sex of human beings and live stocks, embryos fertilized invitro can be determined by PCR by using primers and DNA probes specific for sex chromosomes. PCR technique is also used to detect sexlinked disorders in fertilized embryos.

A technique in which the pattern of colonies growing on a culture plate is copied. A sterile filter plate is pressed against the culture plate and then lifted. Anemophilous plants have the following characteristic features:
(i) The flowers are produced in pendulous, catkin-like or spike inflorescence.
(ii) The axis of inflorescence elongates so that the flowers are brought well above the leaves.
(iii) The perianth is absent or highly reduced.
(iv) The flowers are small, inconspicuous, colourless, not scented, do not secrete nectar.
(v) The stamens are numerous, filaments are long, exerted and versatile.
(vi) Anthers produce enormous quantities of pollen grains compared to a number of ovules available for pollination. They are minute, light and dry so that they can be carried to long distances by wind.
(vii) In some plants, anthers burst violently and release the pollen into the air. Example: Urtica.
(viii) Stigmas are comparatively large, protruding, sometimes branched and feathery, adapted to catch the pollen grains. Generally, a single ovule is present.
(ix) The plant produces flowers before the new leaves appear, so the pollen can be carried without hindrance of leaves.

There are 5 types of ovules.
1. Orthotropous:
Micropyle is at the distal end and the micropyle, the funicle and the chalaza lie in one straight vertical line.
2. Anatropous:
The body of the ovule becomes completely inverted so that the micropyle and funiculus come to lie very close to each other.
E.g: Dicots, Monocots (Common)
3. Hemianatropous:
The body of the ovule is placed transversely and at right angles to the funicle.
E.g: Primulaceae.
4. Campylotropous:
(i) The body of the ovule at the micropylar end is curved and more or less bean shaped. The embryo sac is slightly curved.
(ii) All the three, hilum, micropyle and chalaza are adjacent to one another, with the micropyle oriented towards the placenta.
E.g: Leguminosae.
5. a) Amphitropous:
The distance between hilum and chalaza is less. The curvature of the ovule leads to horse shoe shaped nucellus.
E.g: Alismataceae.
b) Circinotropous:
Funiculus is very long and surrounds the ovule.
E.g: Cactaceae

Autogenic succession
Autogenic succession occurs as a result of biotic factors. The vegetation reacts with its environment and modifies its own environment causing its own replacement by new communities. This is known as autogenic succession.
Example: In forest ecosystem, the larger trees produce broader leaves providing shade to the forest floor area. It affects the shrubs and herbs which require more light (heliophytes) but supports the shade tolerant species (sciophytes) to grow well.
Allogenic succession
Allogenic succession occurs as a result of abiotic factors. The replacement of existing community is caused by other external factors (soil erosion and leaching, etc.,) and not by existing organisms.
Example: In a forest ecosystem soil erosion and leaching alter the nutrient value of the soil leading to the change of vegetation in that area.

Morphological adaptations:
Root system is extensively developed. These roots may be of two types. They are Clinging roots and Aerial roots.
Clinging roots fix the epiphytes firmly on the surface of the supporting objects.
Aerial roots are green coloured roots which may hang downwardly and absorb moisture from the atmosphere with the help of a spongy tissue called velamen.
1. Stem of some epiphytes are succulent and develop pseudo bulb or tuber.
2. Generally the leaves are lesser in number and may be fleshy and leathery.

Seaweed liquid fertilizer (SLF) contains cytokinin, gibberellins, and auxin apart from macro and micro nutrients. Most seaweed-based fertilizers are made from kelp (brown algae) which grows to length of 150 metres. Liquid seaweed fertilizer is not only organic but also eco-friendly. The alginates in the seaweed that reacts with metals in the soil and form long, crosslinked polymers in the soil. These polymers improve the crumbing in the soil, swell up when they get wet and retain moisture for a long time. They are especially useful in organic gardening which provides carbohydrates for plants. Seaweed has more than 70 minerals, vitamins and enzymes. It promotes vigorous growth. Improves resistance of plants to frost and disease. Seeds soaked in seaweed extract germinate much rapidly and develop a better root system.

Radioactive waste management involves the treatment, storage and disposal of liquid, airborne and solid effluents from the nuclear industry. Three ways are employed to manage nuclear wastes
(i) Spent Fuel Pools - The spent fuel discharged from the reactors is temporarily stored in the reactor pool. The Spent fuel rods are used in stored cooling ponds. They protect the surroundings from radiation and absorb the heat generated during radioactive decay.
(ii) Vitrification method - This prevents reaction or degradation of nuclear waste for extended periods of time and encased in dry cement caskets.
(iii) Geological Repositories - A deep geological repository is a nuclear waste repository excavated deep within a stable geologic environment. It is suited to provide a high level of long-term isolation and containment without future maintenance. In India at Tarapur and Kalpakkam, a wet storage facility of Spent Fuel is the main mode of storage.

Sewage treatment is usually performed in the following three stages:
(I) Primary treatment:
(a) Primary treatment involves the physical removal of solid and particulate organic and inorganic materials from the sewage through filtration and sedimentation.
(b) Floating debris is removed by sequential filtration. Then the grit (soil and small pebbles) are removed by sedimentation.
(c) All solids that settle form the primary sludge and the supernatant forms the effluerit and is taken for secondary treatment.
(II) Secondary treatment or biological treatment:
(a) The primary effluent is passed into large aeration tanks where it is constantly agitated mechanically and air is pumped into it.
(b) This allows vigorous growth of useful aerobic microbes into floc (masses of bacteria associated with fungal filaments to form mesh like structures). Which consume the organic matter in the effluent.
(c) This significantly reduces the BOD (Biochemical oxygen demand or Biological oxygen demand). BOD refers to the amount of the oxygen that would be consumed, if all the organic matter in one litre of water were oxidized by bacteria. The sewage water is treated till the BOD is reduced. The greater the BOD of the waste water more is its polluting potential.
(d) Once the BOD of sewage water is reduced Significantly, the effluent is then passed into a settling tank where the bacterial "flocs" are allowed to sediment. Which is called activated sludge. A small part of activated sludge is pumped back into the aeration tank to serve as the inoculum.
(e) The remaining major part of the sludge is pumped into large tanks called anaerobic sludge digesters. Here, the bacteria which grow anaerobically, digest the bacteria and the fungi in the sludge. During digestion, bacteria produce a mixture of gases such as methane, hydrogen sulphide and CO2 Which form biogas and can be used as a source of energy.
(III) Tertiary treatment
(a) Tertiary treatment is the final process that improves the quality of the waste water before it is reused, recycled or released into natural water bodies.
(b) This treatment removes the remaining inorganic compounds and substances, such as nitrogen and phosphorus.
(c) UV is an ideal disinfectant for wastewater since it does not alter the water quality - except for inactivating microorganisms. UV is a chemical-free process that can completely replace the existing chlorination system and also inactivates chlorine-resistant microorganisms like Cryptosporidium and Giardia.

Basic concepts of Tissue Culture
Basic concepts of plant tissue culture are totipotency, differentiation, dedifferentiation and redifferentiation.
Totipotency
The property of live plant cells that they have the genetic potential when cultured in nutrient medium to give rise to a complete individual plant.
Differentiation
The process of biochemical and structural changes by which cells become specialized in form and function.

Redifferentiation
The further differentiation of already differentiated cell into another type of cell. For example, when the component cells of callus have the ability to form a whole plant in a nutrient medium, the phenomenon is called redifferentiation.
Dedifferentiation
The phenomenon of the reversion of mature cells to the meristematic state leading to the formation of callus is called dedifferentiation. These two phenomena of redifferentiation and dedifferentiation are the inherent capacities of living plant cells or tissue. This is described as totipotency.
41.
Direct or Vector less Gene Transfer
In the direct gene transfer methods, the foreign gene of interest is delivered into the host plant without the help of a vector, The following are some of the common methods of direct gene transfer in plants.
(i) Chemical mediated gene transfer: Certain chemicals like polyethylene glycol (PEG) and dextran sulphate induce DNA uptake into plant protoplasts.
(ii) Microinjection: The DNA is directly injected into the nucleus using fine tipped glass needle or micro pipette to transform plant cells. The protoplasts are immobilized on a solid support (agarose on a microscopic slide) or held with a holding pipette under section.
(iii) Electroporation Methods of Gene Transfer: A pulse of high voltage is applied to protoplasts, cells or tissues which makes transient pores in the plasma membrane through which uptake of foreign DNA occurs.

(iv) Liposome mediated method of Gene transfer: Liposomes the artificial phospholipid vesicles are useful in gene transfer. The gene or DNA is transferred from liposome into vacuole of plant cells. It is carried out by encapsulating DNA into the vacuole. This technique is advantageous because the liposome protects the introduced DNA from being damaged by the acidic pH and protease enzymes present in the vacuole. Liposome and tonoplast of vacuole fusion resulted in gene transfer. This process is called lipofection.

(v) Biolistics: The foreign DNA is coated onto the surface of minute gold or tungsten particles (1-3 \(\mu\)m) and bombarded onto the target tissue or cells using a particle gun (also called as gene gun/ micro projectile gun/shotgun) Then the bombarded cells or tissues are cultured on selected medium to regenerate plants from the transformed cells.

1. The sex determining mechanism of humans is XX-XY type.
2. In humans, out of 23 pairs of chromosomes present, 22 pairs are exactly same in both males and females known as autosomes (body).
3. The 23rd chromosome is the sex chromosome. (i.e) Pair of X-chromosome (XX) is present in females and males having one X and one Y chromosome (XY).
4. During, spermatogenesis, in case of males two types of gametes are produced (x and y).
5. The 50% total sperms produced carry X-chromosomes and the rest 50% carry Y - chromosomes besides autosomes. This condition is called male diagamety.
6. The Y containing sperms and X containing sperms are called androsperms and gynosperms respectively.
7. Female produces only one type of ovum with an X-chromosome. If an ovum fertilizes with a sperm carrying X chromosome the zygote develops into a female (XX) and if an ovum is fertilized with sperm carrying Y-chromosome, zygote thus, formed will be male (YY).

8. Hence, the genetic makeup of sperm, which fertilizes the ovum determines the sex of a child there are 50% chances of having either a male or female in each progeny.

(a) Some organisms like honeybees are called parthenogenetic animals.
1. In case of honeybees both haploid and diploid individuals formed, as a result of incomplete parthenogenesis.
2. Both sexual reproduction and parthenogenesis are inclusive.
3. Fertilized eggs (zygote) give rise to queen and workers (both are females) by the process of sexual reproduction.
4. Unfertilized eggs (ova) of females develop into drones (males) by the process of parthenogenesis.
(b) A male honeybee has 16 chromosomes where as its female has 32 chromosomes
1. It is because of haplodiploidy which is a sex determination system, in which males develop from unfertilized eggs (parthenogenesis) and are haploid (16n) and females develop frorn fertilized eggs and are diploid (32n) (sexual reproduction).
2. As unfertilized egg carries only half number of chromosome as compared to fertilized egg.
Therefore male honeybees have 16 chromosomes where as female has 32 chromosomes.
