(a) (Collection of) methods that allows correction of gene defect that has been diagnosed in a child/embryo/Here the genes are inserted into a person's cells and tissues to treat a hereditary disease. It compensate the non-functional gene this involves delivery of a normal gene into the individual/embryo to take over the function of non-functional/a defective gene.
(b) If the desired gene is isolated and introduced into cells at early embryonic stages it can provide a permanant cure. ADA/Adenosine deaminase deficiency

The presence of a chromogenic substrate gives blue coloured colonies, in absence of an insert/in non-transformants, presence of an insert (in the enzyme site), results into (insertional inactivation of the β-galactosidase) colonies which do not produce colour.

Detailed Answer:
Adenosine deaminase deficiency is a genetic disorder. The disorder is caused due to the deletion of the gene for adenosine deaminase, the enzyme crucial for the immune system to function. Adenosine deaminase deficiency in patients can be treated by enzyme replacement therapy. In this treatment, patients are regularly injected with the functional ADA enzyme.
Disadvantages of this process:
(a) It does not completely eradicate the disease.
(b) The requirement of repeated doses of the enzymes makes it expensive.

Drug PTC, 124 is taken orally while gene therapy requires the delivery of gene by vectors into respiratory tract. PTC 124 can be self administered while gene therapy requires proper medical treatment monitored by professionals. PTC 124 is readily taken up by cells while gene by vector system may be poorly or partially taken up by few cells.PTC 124 may have fewer or no side effects while gene therapy has possibility of side effects, i.e. viral vectors may cause infections.

(B) An organism which becomes transformed following the introduction of new DNA sequence into its geneome is called transgenic organism. Most popularly the transgenic crops are called genetically modified crops or GM crops.
A transgenic crop that has been recently introduced in India is Bt cotton. This transgenic variety of cotton contains a foreign gene obtained from a soil bacterium -Bacillus thuringiensis. The bacterium produces a crystal (Cry) protein which is toxic to larvae of certain insects.The gene responsible for production of Cry protein, i.e.., cry gene, has been isolated and introduced into the cotton genome so that the transgenic Bt cotton has become insects resistant.The Bt cotton is now grown in different parts of India and has resulted approximately 3% to 27% increase in cotton yield.
